INTRODUCTION AND HYPOTHESIS: The objective was to investigate the correlation between endogenous vaginal microecological alterations and female pelvic organ prolapse (POP). METHODS: Patients who underwent vaginal hysterectomy were retrospectively analyzed as the POP group (n = 30) and the non-POP group (n = 30). The vaginal microbial metabolites and enzyme levels were tested using the dry chemoenzymatic method. The mRNA and protein expression were tested using real-time quantitative PCR and immunohistochemistry. SPSS version 25.0 and GraphPad Prism 8.0 were performed for statistical analysis. RESULTS: Compared with the non-POP group, the vaginal pH, H2O2 positivity and leukocyte esterase positivity were higher in patients with POP (all p < 0.05). Further analysis showed that patients with pelvic organ prolapse quantification (POP-Q) stage IV had higher rates of vaginal pH, H2O2 positivity and leukocyte esterase positivity than those with POP-Q stage III. Additionally, the mRNA expression of decorin (DCN), transforming growth factor beta 1 (TGF-ß1), and matrix metalloproteinase-3 (MMP-3) in uterosacral ligament tissues were higher, whereas collagen I and III were lower. Similarly, the positive expression of MMP-3 in uterosacral ligament tissue was significantly upregulated in the POP group compared with the non-POP group (p = 0.035), whereas collagen I (p = 0.004) and collagen III (p = 0.019) in uterosacral ligament tissue were significantly downregulated in the POP group. Correlation analysis revealed that there was a significant correlation between vaginal microecology and collagen metabolism. In addition, MMP-3 correlated negatively with collagen I and collagen III (p = 0.002, r = -0.533; p = 0.002, r = -0.534 respectively), whereas collagen I correlated positively with collagen III (p = 0.001, r = 0.578). CONCLUSIONS: Vaginal microecological dysbiosis affects the occurrence of female POP, which could be considered a novel therapeutic option.

The aim of the present study was to investigate the impact and mechanism of high mobility group box 1 (HMGB1) on the regulation of cell migration and invasion in A2780/DDP cisplatin-resistant ovarian cancer cells. After transfecting small interfering (si)RNA-HMGB1 into A2780/DDP cells, Transwell migration and invasion assays were conducted to assess alterations in the cell migratory and invasive abilities. Additionally, western blotting analyses were performed to examine changes in HMGB1, phosphorylated (p)-GSK-3ß, GSK-3ß, E-cadherin and vimentin expression levels. The results of the present study demonstrated that the migratory and invasive abilities of A2780/DDP cells were significantly higher compared with those of A2780 cells. Additionally, the expression levels of HMGB1, p-GSK-3ß and the mesenchymal phenotype marker, vimentin, in A2780/DDP cells were significantly elevated relative to the levels in A2780 cells. Conversely, the expression level of the epithelial phenotype marker, E-cadherin, was markedly decreased compared with that in A2780 cells. Following transfection of A2780/DDP cells with siRNA-HMGB1, there was a significant reduction in the rate of cell migration and invasion. Simultaneously, the expression levels of HMGB1, p-GSK-3ß and vimentin were downregulated while the level of E-cadherin was upregulated. It was therefore concluded that the high expression of HMGB1 in A2780/DDP cells enhanced the cell migration and invasion abilities by facilitating epithelial to mesenchymal transition via GSK-3ß.

Objective: To investigate the clinical characteristics and prognostic factors in patients with endometriosis-associated ovarian cancer. Methods: In this study, we retrospectively analyzed the medical records of 135 ovarian cancer patients admitted to our hospital from January 2016 to January 2018. Based on the presence of concomitant endometriosis (EMs), the patients were divided into two groups: the Endometriosis-Associated Ovarian Cancer (EAOC) group (n=64) and the non-EAOC (NEAOC) group (n=71). We compared the clinical characteristics of the two groups. Additionally, in the EAOC group, we followed up with patients for 5 years, categorized them into the survival group (n=40) and the deceased group (n=24) based on their prognosis, and conducted univariate and multivariate logistic regression analyses to identify influencing factors. Results: In comparison to the NEAOC group, patients in the EAOC group exhibited higher rates of menopause occurrence, pathological stages I-II, vaginal bleeding, and history of cesarean section, with statistical significance (P < .05). They also had a lower incidence of dysmenorrhea, lymph node metastasis, and abdominal distension, as well as an earlier age of onset, all of which were statistically significant (P < .05). There were no statistically significant differences (P > .05) between the two groups in terms of parity, gravidity, tumor diameter, abdominal pain incidence, and body mass index. Based on prognosis, the patients were categorized into a survival group (n=40) and a deceased group (n=24). Comparison between the two groups showed statistically significant differences (P < .05) in terms of postoperative residue, epithelial-mesenchymal transition, and lymph node metastasis. In contrast, there were no statistically significant differences (P > .05) in terms of tumor laterality, histological type, tumor stage, differentiation degree, and vaginal bleeding. The variables with P < .05 were assigned as independent variables, with the prognosis of death as the dependent variable. Multivariate logistic regression analysis revealed that epithelial-mesenchymal transition and lymph node metastasis were independent risk factors for mortality in EAOC patients (P < .05). Conclusion: Clinical characteristics of EAOC patients show significant differences, with epithelial-mesenchymal transition and lymph node metastasis being identified as independent adverse prognostic factors associated with poor outcomes in EAOC patients. However, this study has limitations such as a relatively small sample size, and further research is therefore necessary.

Chromium (Cr), total arsenic (As), inorganic arsenic (iAs), cadmium (Cd), mercury (Hg), methylmercury (MeHg), and lead (Pb) were analyzed in in Agaricus blazei, Tricholoma matsutake, Pholiota nameko, agrocybe aegirit, Boletus edulis, Auricularia auricula, and Lentinus edodes collected from online supermarket in China from 2015 to 2017. The order of mean concentrations for the five heavy metals in edible mushrooms was As > Cd > Cr > Pb > Hg. No positive correlation was found between total As and iAs, nor between total Hg and MeHg. The contents of iAs were at a low level except for A. blazei samples. The contents of MeHg were at a low level in all test mushroom samples. And Cr, Cd, and Pb pollution were common problems in the test mushroom samples. The comprehensive factor pollution index was between 0.569 (A. auricula) and 3.056 (B. edulis). The THQ values for the five heavy metals from P. nameko, A. auricula, A. aegirit, and L. edodes samples were less than 1. The hazard index (HI) values of A. blazei, T. matsutake, and B. edulis samples for adults and children were greater than 1, indicating significant health hazard to the adults and children consumers. The cancer risk (CR) values for iAs ranged from 3.82 × 10- 6 (T. matsutake) to 8.61 × 10- 5 (A. blazei), indicating no potential carcinogenic risk to the consumers. The order for carcinogenic risk of each edible mushroom species was A. blazei > L. edodes > P. nameko > A. aegirit > A. auricula > B. edulis > T. matsutake.

Background: Long non-coding RNAs (lncRNAs) have been confirmed to play vital roles in tumorigenesis. LncRNA MYU has recently been reported as an oncogene in several kinds of tumors. However, MYU's expression status and potential involvement in ovarian cancer (OC) remain unclear. In this study, we explored the underlying role of MYU in OC. Methods and results: The expression of MYU was upregulated in OC tissues, and MYU's overexpression was significantly correlated with the FIGO stage and lymphatic metastasis. Knockdown of MYU inhibited cell proliferation in SKOV3 and A2780 cells. Mechanistically, MYU directly interacted with miR-6827-5p in OC cells; HMGA1 is a downstream target gene of miR-6827-5p. Furthermore, MYU knockdown increased the expression of miR-6827-5p and decreased the expression of HMGA1. Restoration of HMGA1 expression reversed the influence on cell proliferation caused by MYU knockdown. Conclusion: MYU functions as a ceRNA that positively regulates HMGA1 expression by sponging miR-6827-5p in OC cells, which may provide a potential target and biomarker for the diagnosis or prognosis of OC.

Arsenic (As) is enriched in wild edible fungi, which is one of the main important sources of As in humans' diet. In this study, two wild edible fungi were employed for investigation: (1) Pleurotus citrinopileatusone, which contains a high content of inorganic As (iAs) and (2) Agaricus blazei Murill, which contains a high content of organic As. This study investigated the changes in As content and its speciation after different daily cooking methods. We found that the content of As in Pleurotus citrinipileatus and Agaricus blazei Murill reduced by soaking plus stir-frying by 55.4% and 72.9%, respectively. The As content in Pleurotus citrinipileatus and Agaricus blazei Murill decreased by 79.4% and 93.4%, respectively, after soaking plus boiling. The content of As speciation in dried wild edible fungi reduced significantly after different treatments. Among them, iAs decreased by 31.9~88.3%, and organic As decreased by 33.3~95.3%. This study also investigated the bioaccessibility of As in edible fungi after different cooking processes via an in-vitro physiologically based extraction test (PBET). The results showed that the bioaccessibility of As was relatively high if the edible fungi were uncooked, boiled, or stir-fried. The gastric (G) bioaccessibility of As ranged from 51.7% to 93.0% and the gastrointestinal (GI) bioaccessibility of As ranged from 63.5% to 98.1%. Meanwhile, the bioaccessibility of inorganic As was found to be as high as 94.6% to 151%, which indicates that further evaluation of the potential health risks of wild edible fungi is necessary.

OBJECTIVE: To understand the distribution characteristics of 46 elements in drinking water in Ankang area of Shanxi Province. METHODS: A total of 46 elements in drinking water samples collected in Ankang area during dry and wet seasons in 2020 were determined by inductively coupled plasma mass spectrometry(ICP-MS). According to the "drinking water hygiene standards"(GB 5749-2022) and "food safety national standards". the 46 elements were classified As general chemical indexes(Al, Mn, Fe, Cu, Zn), toxicological indexes(Cr, As, Cd, Hg, Pb, Ba, B, Mo, Ni, Sb, Be, Ag, Tl), new reference indexes(U, V), and major elements(K, Ca, N) A, Mg), trace elements(Li, Co, Se, Sr, Sn) and rare earth elements(Sc, Y, La, Ce, Pr, Nd, Sm, Eu, Gd, Tb, Dy, Ho, Er, Tm, Yb, Lu, Th) were analyzed and described. RESULTS: The maximum values of Al and Fe in drinking water in Ankang area were 1.21 and 0.98 mg/L, exceeding the limits of 0.2 and 0.3 mg/L. In dry season, the median content of Fe in drinking water of different water sources was higher in groundwater than in surface water. The Al and Fe of surface water were higher than that of groundwater in wet season. The toxicological indexes all met the standard requirements, and there was no significant difference among districts and counties. The median content of Na in drinking water of different water sources was higher in groundwater than in surface water, while Mg, K and Ca were higher in surface water than in groundwater. The maximum value of the newly added reference index U was 0.015 mg/L lower than the limit standard 0.03 mg/L, and the maximum value of V was 0.019 mg/L higher than the limit standard 0.01 mg/L, but the median of both indexes were low. The median content of Li in drinking water of different water sources was that surface water was higher than groundwater, while Se and Sr were higher in groundwater than surface water. The maximum content of Se was 0.016 mg/L in Ziyang County, and Sr content was generally higher. The content of rare earth elements was low, mostly below the detection. CONCLUSION: The drinking water quality in Ankang area is excellent and rich in strontium. However, the general chemical indexes Al, Fe and the newly added appendix index V in some sampling points exceed the limit standard, so supervision and monitoring should be strengthened.

The studies on how humic acid (HA) influences the oxidative stress of arsenic in aquatic organism is limited. Using Danio rerio as case study, we explored the oxidative stress effects in aquatic organism after 96 h exposure to the HA and arsenic. Results revealed the co-exposure of HA and arsenite elevated the superoxide dismutase activities and downgraded the malondialdehyde. Thus, we speculate that HA may alleviate the oxidative stress induced by arsenite, which may be caused by the HA's coating in combination with the complexation of arsenite and HA. In addition, HA acted as the reactive oxygen species scavenger, promising to eliminate the oxygen free radicals. Contrastingly, HA may impact little on the arsenate exposure. This study can help better understand oxidative stress mechanism of co-exposure of arsenic and HA in aquatic environment.

A method for the simultaneous determination of arsenobetaine, arsenite, arsenate, dimethylarsinic acid, monomethylarsonic acid, selenite, selenate, bromate, bromide, iodate, and iodide in bottled drinking water and fruit juice samples was established by using high-performance liquid chromatography-inductively coupled plasma mass spectrometry. The separation of eleven compounds was performed on an ion exchange chromatography column (Dionex IonPac AS14) with 20 mmol L-1 (NH4)2CO3 (pH 10) and 50 mmol L-1 (NH4)2CO3 (pH 10) as a mobile phase. The limits of quantification of the method were 0.17 - 1.2 µg L-1 for the test compounds in bottled drinking water and 0.34 - 2.4 µg L-1 in fruit juice. The average recoveries ranged from 85.8 to 102.2%, and the relative standard deviations (RSDs) obtained in fortification recovery studies were generally <4.2% for bottled drinking water samples. The average recoveries ranged from 88.1 to 118.0% (except for iodate) for fruit juice sample.

PURPOSE: To determine the risk factors of intraoperative cyst rupture in partial nephrectomy (PN) for a cystic renal mass (CRM) and their effect on the prognosis of patients. MATERIALS AND METHODS: Patients who underwent partial nephrectomy for CRMs from January 2009 to January 2015 were included. Uni/multivariate Logistic/Cox analysis and Kaplan-Meier analysis were performed. RESULTS: A total of 174 patients were included in this study. There were 27 (15.5%) intraoperative cyst ruptures. The median follow-up time was 60 months. Multivariate logistic analysis showed that the E component (P = 0.018) and N component (P = 0.022) of the R.E.N.A.L. nephrometry score, Bosniak category III (P = 0.044), and surgeon's experience (P = 0.030) were risk factors associated with intraoperative cyst rupture in PN for CRMs. The 5-year recurrence-free survival (RFS), cancer-free survival (CFS) and overall survival (OS) were 92.7%, 90.32% and 94.4%, respectively, in 124 cases of malignant CRM. Kaplan-Meier analysis demonstrated that 5-year RFS and 5-year CFS in patients with cyst rupture was worse than those without cyst rupture (P = 0.006 and 0.003, respectively). Multivariate Cox analysis revealed that intraoperative cyst rupture was independent risk factor for 5-year RFS and 5-year CFS (P = 0.039 and 0.013, respectively). However, there was no significant difference in OS between the two groups (P = 0.275). CONCLUSIONS: The prevalence of intraoperative cyst rupture is relatively high. Higher E and N scores, Bosniak category III, and lacking surgical experience (<20 cases) increase the risk of occurrence of intraoperative cyst rupture.

PURPOSE: To explore the exact mechanism through which emodin down-regulates the migration and invasion abilities of endometrial stromal cells. Moreover, to explore the theoretical basis of emodin in the treatment of endometriosis. PATIENTS AND METHODS: Endometriosis endometrial stromal cells (EESs) were cultured from 15 women with endometriosis and control endometrial stromal cells (CESs) were cultured from 12 women without endometriosis. The levels of proteins were evaluated by Western blot. The migration and invasion abilities of cells were detected by transwell assays. RESULTS: The abilities of migration and invasion of EESs were much stronger than those of CESs. After treated with emodin, the migration and invasion abilities of EESs and CESs were significantly down-regulated, and the levels of integrin-linked kinase (ILK) and p-GSK-3ß were statistically down-regulated in EESs. Besides that, the expression of keratin was up-regulated while the expression of vimentin, ß-catenin and slug were all down-regulated by emodin in a dose- and time-dependent manner. Silencing of ILK gene in EESs also achieved the above effects, which were strengthened by emodin. Conversely, exogenous expression of ILK in CESs increased the expression of p-GSK-3ß, which were abrogated by emodin. Furthermore, SB216763 increased migration and invasion abilities of CESs by facilitating the epithelial-mesenchymal transition (EMT) through up-regulating levels of p-GSK-3ß, ß-catenin and slug, which were also abrogated by emodin. CONCLUSION: Emodin inhibits the migration and invasion abilities of human endometrial stromal cells by reversing the EMT via ILK/GSK-3ß pathway. So, emodin may be considered as a promising targeted therapy for endometriosis.

Cardamine violifolia (Brassicaceae) is a novel selenium(Se) hyperaccumulation plant with rich nutrients, and serves as a good source of special vegetables in Enshi, China. The present study aimed to investigate the effects of the application of selenate, selenite, and Se yeast (50-800 mg/L) on the growth, Se accumulation, nutrient uptake, and antioxidant response of C. violifolia. The results showed that the Se accumulation efficiency was selenate > selenite > Se yeast, the maximum Se concentration could achieve over 7000 mg/kg, and about 90% was organic Se. The major Se speciation found was mainly SeCys2 and the proportion of various Se species were affected by the Se forms and concentrations. Besides, the plant growth, nutrition quality indexes, element uptakes, and antioxidant responses indicated that 200 mg/L selenate was optimum for C. violifolia to accumulate Se without much impacts, while to obtain more proportion of organic Se, 200 mg/L selenite might be a better choice.

Purpose: To investigate the risk factors for progression to castration-resistant prostate cancer (CRPC) in metastatic prostate cancer (mPCa) patients who underwent androgen deprivation therapy (ADT). Methods: We analyzed 216 patients with mPCa who underwent ADT between January 2006 and December 2015 at the First Affiliated Hospital of Fujian Medical University. Univariate and multivariate Cox regression analysis were used to explore the risk factors for progression to CRPC. Kaplan-Meier analysis and log-rank test were used to evaluate the difference in progression-free survival (PFS). Results: A total of 121 (56.0%) patients who underwent ADT showed progression to CRPC. Multivariate Cox regression analysis demonstrated that Gleason grade group, prostate-specific antigen nadir (nPSA), and time to PSA nadir (TTN) were risk factors for progression to CRPC in mPCa patients. Kaplan-Meier analysis demonstrated that patients in Gleason grade group ≥3, nPSA >0.2 ng/ml and TTN <6 months had shorter PFS. Conclusion: This study demonstrated that Gleason grade group, nPSA and TTN were risk factors for progression to CRPC. Patients with higher Gleason grade group, higher nPSA and shorter TTN have shorter PFS and higher risk of progression to CRPC after ADT.

RESUMEN

OBJECTIVE: To investigate the influence of the degrees of intravesical prostatic protrusion (IPP) on the recovery of urinary continence after radical prostatectomy. METHODS: We retrospectively analyzed the clinical data on 212 patients diagnosed with prostate cancer by biopsy and treated by laparoscopic radical prostatectomy by the same surgeon. Based on the degrees of IPP measured by MRI, we divided the patients into an IPP ≤ 10 mm group (n = 146) and an IPP > 10 mm group (n = 66) and determined the factors influencing the recovery of urinary continence by univariate and multivariate logistic regression analyses. RESULTS: At 1, 3, 6 and 12 months after surgery, the urinary continence rates of the patients were 32.5%, 50.5%, 82.1% and 91%, respectively. Univariate analysis indicated that the factors influencing the recovery of urinary continence included IPP, body mass index (BMI), bladder neck preservation (BNP), neurovascular bundle preservation (NVBP) and clinical tumor (T) stage at 3 months (P < 0.05 or P < 0.01), age, IPP, BMI, BNP and clinical T stage at 6 months (P < 0.05 or P < 0.01), and age, IPP, BMI, BNP, NVBP and clinical T stage at 12 months (P < 0.05), while multivariate logistic regression analysis showed the independent influencing factors to be IPP > 10 mm (P < 0.001), BMI ≥ 25 kg/m2 (P = 0.004) and BNP (P = 0.032) at 3 months, and IPP and BMI at 6 months (both P < 0.01) and 12 months (P < 0.01 and P = 0.033). CONCLUSIONS: IPP > 10 mm and BMI ≥ 25 kg/m2 are independent factors influencing the long-term recovery of urinary continence after radical prostatectomy.

RESUMEN

OBJECTIVE: A method was established to determine mercury in seven sorts of disinfectant by water-bath-microwave digestion-atomic fluorescence spectrometry. METHODS: The samples of disinfectant were preliminary digestion by water-bath at 90℃and then dissolved by microwave according to the temperature programmed. The mercury ions in the digested solution reacted with reductant to generate the mercury vapour which were measured the fluorescence intensity by the atomic fluorescence spectrometer. RESULTS: The linear range was from 0- 15 µg / L and the correlation coefficient was 0. 9995. The detection limit of this method was 0. 17 µg / kg. The precision of the method was less than4. 2% and the spiked recoveries of high, medium and low concentrations were between87%- 105%. CONCLUSION: This method possesses several advantages including less interference, high accuracy, good reproducibility and high sensitivity. It is suitable for the current determination of mercury in the seven sorts of disinfectant.
